羧甲基纤维素钠(CMC)是一种纤维素的衍生物,在食品、日化、医药及化工等领域有广泛的应用。为进一步研究羧甲基纤维素钠在生物体内的药物代谢性质,开发以羧甲基纤维素钠为载体的体内药物,本项目进行了14C标记羧甲基纤维素钠的制备研究。以碱化纤维素为原料,14C-氯乙酸与纤维素上羟基通过醚化反应实现14C标记羧甲基纤维素钠的制备。详细研究了碱化纤维素与氯乙酸用量比例、醚化反应时间、反应温度对标记反应的影响以及标记物的纯化方法,确定了最佳标记反应条件。应用优化后的标记条件成功制备了7.92×107 Bq
Sodium carboxymethyl cellulose (CMC) is a cellulose derivative that is widely used in food, cosmetic, pharmaceutical and chemical industries. In order to further study the pharmacokinetics of sodium carboxymethylcellulose in vivo and to develop an in vivo drug with sodium carboxymethylcellulose as carrier, 14C-labeled sodium carboxymethylcellulose was prepared in this project. Preparation of 14C-labeled sodium carboxymethylcellulose with 14C-chloroacetic acid and hydroxyl groups on cellulose by etherification with alkalized cellulose as raw material. The effects of the ratio of alkali cellulose and chloroacetic acid, the reaction time of etherification, the effect of reaction temperature on the labeling reaction and the purification method of the label were studied in detail. The optimum labeling reaction conditions were determined. 7.92 × 107 Bq was successfully prepared using the optimized labeling conditions
