目的:观察核受体相关因子1(nuclear receptor-related factor 1,Nurr1)与多巴胺能神经细胞特异性标记物酪氨酸羟化酶(TH)在生后大鼠视网膜发育过程中的表达变化,探明Nurr1与视网膜多巴胺能神经元的相关关系。方法:石蜡包埋组织切片,免疫组织化学双重标记。结果:Nurr1在视网膜发育过程中的表达出现了显著的动态变化,Nurr1阳性产物主要表达在内核层细胞,生后3~7 d达到高峰,之后随着细胞的成熟阳性表达又逐渐减少,成熟的视网膜组织内仅见少量Nurr1阳性细胞,在视网膜神经细胞从幼稚到成熟的分化过程中仅见个别TH阳性细胞。Nurr1与TH的免疫组化双标结果显示两蛋白可以共表达在同一细胞中,但众多的Nurr1阳性细胞不表达TH。结论:Nurr1在大鼠视网膜多巴胺能神经细胞及非多巴胺能神经细胞从幼稚到成熟的分化过程中可能具有重要的调控作用。
Objective: To observe the expression changes of nuclear receptor-related factor 1 (Nurr1) and dopaminergic neuron specific marker tyrosine hydroxylase (TH) during retina development in postnatal rats, To explore the relationship between Nurr1 and retinal dopaminergic neurons. Methods: Paraffin-embedded tissue sections were double-labeled with immunohistochemistry. RESULTS: The expression of Nurr1 in retina was significantly different from that in the control group. The positive expression of Nurr1 was mainly expressed in the inner nuclear layer cells, reaching the peak at 3 ~ 7 d after birth, and then gradually decreased with the maturation of the cells. The mature Only a small amount of Nurr1 positive cells were seen in the retina, and only a few TH positive cells were seen during the process of differentiation of retinal neurons from immature to mature. Immunohistochemistry with Nurr1 and TH double-labeled results showed that the two proteins can be co-expressed in the same cell, but a large number of Nurr1-positive cells do not express TH. Conclusion: Nurr1 may play an important regulatory role in the process of retinal dopaminergic neurons and non-dopaminergic neurons from immature to mature.
