目的建立三角帆蚌中主要核苷类成分尿嘧啶、次黄嘌呤、黄嘌呤、尿苷、胸腺嘧啶的HPLC测定方法。方法采用Lichrosper-C_(18)柱,以水为流动相洗脱,检测波长254 nm,体积流量为1.0 mL·min~(-1),柱温30℃。结果三角帆蚌中尿嘧啶、次黄嘌呤、黄嘌呤、尿苷和胸腺嘧啶的含量分别为153.2~173.3μg·g~(-1)、106.6~360.8μg·g~(-1)、117.6~172.8μg·g~(-1)、70.3~153.1μg·g~(-1)和343.9~511.4μg·g~(-1)。结论本方法简便、准确、重现性好,可作为三角帆蚌中主要核苷类成分含量的常规检测方法。
Objective To establish a HPLC method for the determination of uridine, hypoxanthine, xanthine, uridine, and thymine, the main nucleosides in the spinnaker. Methods Lichrosper-C_(18) column was used with water as the mobile phase. The detection wavelength was 254 nm, the volumetric flow rate was 1.0 mL·min -1, and the column temperature was 30°C. Results The contents of uracil, hypoxanthine, xanthine, uridine and thymidine in swiftlet were 153.2~173.3μg·g -1, 106.6~360.8μg·g -1, and 117.6~ 172.8 μg·g -1, 70.3 - 153.1 μg·g -1, and 343.9 - 511.4 μg·g -1. Conclusion This method is simple, accurate, and reproducible. It can be used as a routine detection method for the content of major nucleosides in the magnolia.
