目的:探讨急性期2肾1夹(2K1C)高血压主动脉单核细胞趋化因子-1(MCP-1)的表达以及氯沙坦 (losartan)干预对其影响。方法:采用银夹法复制2K1C高血压模型,用放免法测定血浆和主动脉壁血管紧张素ⅡAngⅡ)浓度,用原位杂交和RT-PCR方法检测2K1C高血压大鼠在血压升高不同时间以及氯沙坦干预28 d后主动脉MCP-1 mRNA的表达。结果:正常血压大鼠主动脉无MCF-1的表达;在2K1C大鼠高血压形成1周,主动脉MCP-1即有表达,并随时间延长,其表达持续增加;氯沙坦干预28 d后,2K1C高血压大鼠主动脉MCP-1的表达明显减少,差异有统计学意义;2K1C高血压大鼠血浆和主动脉壁AngⅡ浓度增加,但只有主动脉壁局部的 AngⅡ浓度同MCP-1表达的吸收度值呈正相关。结论:急性期2K1C高血压大鼠主动脉即有MCP-1的表达并随时间延长而增加,氯沙坦干预可明显减少其表达。
Objective: To investigate the expression of MCP-1 in acute 2K1C 2K1C hypertensive aorta and the effect of losartan on it. Methods: The model of 2K1C hypertension was replicated by silver clip method. The concentration of angiotensin Ⅱ AngⅡ in plasma and aortic wall was determined by radioimmunoassay. The in situ hybridization and RT-PCR were used to detect the changes of blood pressure in 2K1C hypertensive rats at different times and Losartan intervention 28 d after aortic MCP-1 mRNA expression. Results: The expression of MCF-1 was not detected in the aorta of rats with normal blood pressure. MCP-1 expression was observed in the aorta of rats with 2K1C for 1 week. The expression of MCP-1 increased with the prolongation of time. , The expression of MCP-1 in the aorta of 2K1C hypertensive rats decreased significantly, the difference was statistically significant; AngⅡ concentration in plasma and aorta increased in 2K1C hypertensive rats, but only AngⅡ concentration in the aorta wall was the same as MCP-1 The expression of the absorption value was positively correlated. CONCLUSION: The expression of MCP-1 in the aorta of 2K1C hypertensive rats in acute stage increases with the prolongation of time. Losartan treatment can significantly reduce the expression of MCP-1.
