目的检测黄绿青霉素对心肌细胞DNA的损伤。方法心肌细胞原代培养和单细胞凝胶电泳法。结果13.0μmolL黄绿青霉素处理组心肌细胞发生凋亡,凋亡率为35.6%,凋亡细胞的TailDNA%、LH、ExtentTM、TailLength4项指标均明显高于其他剂量组,差异显著;而2.6、1.3、0.625μmolL黄绿青霉素处理组,未观察到明显的拖尾现象,以上4项指标与对照组相比无明显变化。结论13μmolL黄绿青霉素具有致心肌细胞DNA损伤的作用。
Objective To detect the DNA damage of cardiomyocytes induced by penicillin. Methods Primary culture of cardiomyocytes and single cell gel electrophoresis. Results Apoptosis of cardiomyocytes was observed in 13.0μmolL xanthrinomycin group with a apoptotic rate of 35.6%. TailDNA%, LH, ExtentTM and TailLength4 of apoptotic cells were significantly higher than those of other groups , 0.625μmolL yellow penicillin treatment group, no significant trailing phenomenon was observed, the above four indicators no significant change compared with the control group. Conclusion 13μmolL xanthogenin can induce DNA damage in cardiomyocytes.
