目的:观察长期使用L-精氨酸(L-NNA)和谷氨酸单氨钠(MSG)后高血压大鼠血浆血管紧张素II(angiotensinII,ANGII)的变化。方法:建模后每日腹腔注射L-NNA和MSG,于7、30、70d用放免法测定大鼠血浆ANGII的质量浓度。结果:(1)高血压组各时期的血压均明显高于对照组(P<0 05);注射L-NNA组的血压在7d与高血压组无明显差异,但30、70d显著增高(P<0 05);注射MSG组的血压显著低于高血压组和注射L-NNA组,但仍未恢复至正常水平。(2)对照组大鼠各时期血浆ANGII的质量浓度介于(162 6±49 1)与(181 5±37 8)μg/L之间,高血压组大鼠血浆ANGII的质量浓度均高于对照组,7d时无差异,但30、70d差异显著(P<0 05);注射L-NNA后,ANGII的质量浓度升高明显,其中30、70d具有显著差异(P<0 05),而注射MSG组大鼠血浆ANGII的质量浓度明显降低(P<0 05),但仍然高于对照组和高血压组(P<0 05)。结论:一氧化氮可有效的减少外周组织ANG的分泌,其降压作用可能通过有效的降低血液中ANG的质量浓度来实现。
Objective: To observe the changes of plasma angiotensin II (ANGII) in hypertensive rats after prolonged use of L-arginine (L-NNA) and glutamate monosodium glutamate (MSG). Methods: After intraperitoneal injection of L-NNA and MSG, the concentrations of ANGII in rat plasma were determined by radioimmunoassay at 7, 30 and 70 days after modeling. Results: (1) The blood pressure in hypertension group was significantly higher than that in control group at each time point (P <0.05). The blood pressure in L-NNA group was not significantly different from that in hypertension group at 7 days, but significantly increased at 30 and 70 days <0.05). The blood pressure of MSG group was significantly lower than that of hypertension group and L-NNA group, but it did not return to normal level. (2) The concentrations of ANGII in plasma of rats in control group were (162 6 ± 49 1) and (181 5 ± 37 8) μg / L respectively, and the levels of plasma ANGII in hypertension group were higher than In the control group, there was no difference at the 7th day but significant difference at 30 and 70 days (P <0.05). After injection of L-NNA, the concentration of ANGII increased significantly, with a significant difference at 30 and 70 days (P <0.05) The plasma concentrations of ANGII in MSG group were significantly decreased (P <0.05), but still higher than those in control group and hypertension group (P <0.05). CONCLUSION: Nitric oxide can effectively reduce the secretion of ANG in peripheral tissues, and its antihypertensive effect may be achieved by effectively lowering the concentration of ANG in the blood.
