目的探讨大鼠非梗死区心肌Dock1蛋白的表达改变。方法建立SD大鼠实验性心肌梗死(22只)及假手术模型(20只)。分别于术后24 h和12周时处死心肌梗死组和假手术组大鼠(各8只),获取心脏标本,应用免疫印迹法检测非梗死区心肌及假手术组心肌Dock1蛋白的表达。结果术后24 h时,心肌Dock1蛋白表达在梗死组(0.13±0.03)与假手术组(0.10±0.04)间的差异无统计学意义(P>0.05);但术后12周时,梗死组(0.17±0.04)较假手术组(0.11±0.05)表达增加,差异有统计学意义(P<0.05);术后12周时的梗死组心肌Dock1蛋白表达较24 h时的梗死组和假手术组均增加,差异有统计学意义(P<0.05)。结论心肌中存在Dock1蛋白的表达;梗死后12周时非梗死区心肌Dock1蛋白表达增加。
Objective To investigate the expression of Dock1 protein in non-infarcted myocardium in rats. Methods SD myocardial infarction (22 rats) and sham operation model (20 rats) were established. Myocardial infarction group and sham operation group (8 rats each) were sacrificed at 24 h and 12 weeks after operation respectively. Cardiac specimens were obtained and the expression of Dock1 protein in non-infarcted myocardium and sham operation group was detected by Western blotting. Results There was no significant difference in the expression of Dock1 protein between myocardial infarction group (0.13 ± 0.03) and sham-operated group (0.10 ± 0.04) at 24 h after operation (P> 0.05) (0.17 ± 0.04) compared with the sham operation group (0.11 ± 0.05), the difference was statistically significant (P <0.05). The expression of Dock1 protein in myocardial infarction group at 12 weeks after operation was significantly higher than that in the infarction group and sham operation at 24 hours Group were increased, the difference was statistically significant (P <0.05). Conclusion The expression of Dock1 protein exists in myocardium. The expression of Dock1 protein in non-infarcted myocardium increased at 12 weeks after infarction.
