[目的]研究Gαq对H2O2诱导的人单核细胞白血病细胞(THP-1)凋亡的影响。[方法]实验分为3组:对照组,H2O2诱导凋亡组,Gp-2A抑制组,每组3个重复。Gp-2A抑制组在培养基加入10μM Gp Antagonist 2A(Gαq抑制剂Gp-2A),培养24h后与H2O2诱导凋亡组同时加入等浓度H2O2,对照组加等量PBS。8h后用流式细胞仪检测细胞凋亡。[结果]在1mM H2O2下,加入Gp-2A的实验组凋亡明显低于不加组。[结论]Gp-2A可以明显抑制低浓度下H2O2诱导的Thp-1细胞的凋亡。
[Objective] To investigate the effect of Gαq on H2O2-induced apoptosis of human monocytic leukemia cells (THP-1). [Methods] The experiment was divided into 3 groups: control group, H2O2-induced apoptosis group, Gp-2A inhibition group, 3 replicates in each group. The Gp-2A inhibition group was treated with 10μM Gp Antagonist 2A (Gαq inhibitor Gp-2A) in culture medium for 24 h, then added with H2O2 at the same concentration as H2O2-induced apoptosis group, and the same volume of PBS in control group. After 8h, apoptosis was detected by flow cytometry. [Results] Under the condition of 1 mM H2O2, the apoptosis of the experimental group which added Gp-2A was obviously lower than that of the untreated group. [Conclusion] Gp-2A can significantly inhibit H2O2-induced Thp-1 cell apoptosis at low concentration.
