目的 :研究雷公藤红素 (Tripterine)对狼疮小鼠肾小球硬化的防治作用。 方法 :在不同发病时间、用不同剂量的雷公藤红素对BWF1小鼠进行腹腔注射。每 2周测鼠 2 4h尿蛋白量及血清抗dsDNA抗体水平。 2 0周后检测鼠肾组织Ⅲ型胶原 (collagentypeⅢ ,ColⅢ )、层粘素 (laminin ,LM)、基质金属蛋白酶 (MMP) 1及其组织抑制剂 (TIMP) 1、转化生长因子 (transforminggrowthfactor,TGF) β1mRNA的表达。 结果 :①雷公藤红素抑制小鼠蛋白尿及血清抗dsDNA抗体水平 ,增加肾局部MMP 1而减少ColⅢ、LM、TIMP 1及TGF β1mRNA的表达 ,改善鼠肾组织学病变。②蛋白尿发生前使用雷公藤红素较蛋白尿发生后使用疗效更显著。③蛋白尿发生前使用雷公藤红素 3mg/ (kg·wk)和 6mg/ (kg·wk)的疗效无统计学差异。 结论 :雷公藤红素对该狼疮模型的肾小球硬化具有保护作用。其降低肾脏ColⅢ及LM沉积可能是通过增加小鼠肾局部MMP 1而抑制TGF β1及TIMP 1的表达而实现的
Objective: To study the prevention and treatment of tripterine on glomerulosclerosis in lupus mice. Methods: BWF1 mice were injected intraperitoneally with different doses of tripterine at different time of onset. The quantity of urinary protein and the level of serum anti-dsDNA antibody in 24 h were measured every 2 weeks. Twenty weeks later, the expressions of collagen Ⅲ, laminin (LM), matrix metalloproteinase (MMP) 1 and tissue inhibitor of metalloproteinase 1 (TIMP) 1, transforming growth factor (TGF) ) β1 mRNA expression. Results: Tripterine inhibited mouse proteinuria and serum anti-dsDNA antibody levels, increased renal MMP 1 and decreased the expression of Col Ⅲ, LM, TIMP 1 and TGF β 1 mRNA, and improved renal histological changes. ② the use of triptolide before the occurrence of proteinuria than proteinuria after the use of more significant effect. ③ There was no significant difference in the efficacy of triptolide between 3 mg / (kg · wk) and 6 mg / (kg · wk) before proteinuria. Conclusion: Tripterine has a protective effect on glomerulosclerosis in this lupus model. The decrease of Col Ⅲ and LM deposition in kidneys may be caused by increasing the expression of TGFβ1 and TIMP-1 by increasing the renal MMP 1 in the kidney
