目的 探讨氧化苦参碱对大鼠肝星状细胞增殖的影响及其抗肝纤维化的机理。方法 用链酶蛋白酶和胶原酶原位灌流消化正常大鼠肝脏 ,Nycodenz密度梯度离心分离肝星状细胞 ,以MTT比色法观察氧化苦参碱对肝星状细胞毒性作用 ,3H TdR法观察氧化苦参碱对肝星状细胞增殖的效应。结果 氧化苦参碱浓度 >10 -5mol/l时对肝星状细胞增殖有抑制作用 (P <0 .0 5 )。结论 氧化苦参碱可抑制肝星状细胞的增殖 ,有抗肝纤维化的作用。
Objective To investigate the effect of oxymatrine on the proliferation of rat hepatic stellate cells and the mechanism of anti-hepatic fibrosis. Methods The livers of normal rats were digested in situ with enzyme-protease and collagenase. The hepatic stellate cells were isolated by Nycodenz density gradient centrifugation. The toxic effects of oxymatrine on hepatic stellate cells were observed by MTT colorimetry. The oxidation was observed by 3H TdR method. The effect of matrine on the proliferation of hepatic stellate cells. Results The concentration of oxymatrine >10 -5 mol/l inhibited the proliferation of hepatic stellate cells (P < 0.05). Conclusion Oxymatrine inhibits the proliferation of hepatic stellate cells and has anti-fibrotic effects.
