目的了解异常蛋白负荷对鼠肾小管上皮细胞 CD5 4(ICAM- 1)表达的影响及细胞骨架蛋白的中间丝波形蛋白 (vimentin)表达的影响。方法用 Ig G(10 m g/ m L )和不同浓度的牛血清白蛋白 (BSA) (1,15 ,30 mg/ m L )作用于体外培养的鼠肾小管上皮细胞株 (NRK 5 2 E) ,采用流式细胞仪测定细胞表面 CD5 4的平均荧光强度的变化 ;对 Ig G和 BSA刺激下的爬片细胞用免疫组化的方法检测 vimentin表达的变化。结果 Ig G能引起 CD5 4表达上调 40 .1% (P<0 .0 5 ) ,BSA(15 ,30 m g/ m L )能引起 CD5 4表达上调 2 4.8%和 2 5 .0 % ,但无统计学意义 ;Ig G和 BSA(15 ,30 m g/ m L )可刺激肾小管上皮细胞表达 vim entin(P<0 .0 5 )。结论异常蛋白负荷可诱导肾小管上皮细胞粘附分子表达上调和细胞骨架的变化 ,从而可能是导致肾小管间质炎性浸润和瘢痕化的重要因素之一。
Objective To investigate the effects of abnormal protein load on the expression of CD54 (ICAM-1) in rat renal tubular epithelial cells and the expression of vimentin in cytoskeletal proteins. Methods Rat renal tubular epithelial cells (NRK 5 2 E) cultured in vitro were treated with Ig G (10 mg / mL) and different concentrations of BSA (1, 15, 30 mg / mL) The changes of the average fluorescence intensity of CD5 4 on the cell surface were measured by flow cytometry. The changes of vimentin expression were detected by immunohistochemistry on the glial cells stimulated by Ig G and BSA. Results Ig G up-regulated the expression of CD5 4 by 40.1% (P <0.05) and raised the expression of CD5 4 by 24.8% and 25.5% at BSA (15,30 mg / ml) Statistical significance; Ig G and BSA (15, 30 mg / m L) stimulated renal tubular epithelial cells expressing vim entin (P <0.05). Conclusion Abnormal protein load can induce the up-regulation of renal tubular epithelial cell adhesion molecule expression and cytoskeleton changes, which may be one of the important factors leading to tubulointerstitial inflammatory infiltration and scarring.
