重型乙型病毒性肝炎的病例,血清病毒水平极低,以至于在部分病例即使以极灵敏的聚合酶链反应也难以检测出来。巢式多聚酶链反应可检测出低至1—10个模板DNA的水平,我们应用此方法检测了12例重症肝炎病人血清乙肝病毒DNA。结果:以C_1、C_3引物一次能检出HBVDNA者4例,其中有3例HBeAg阳性者全部被检出;经C_(12)、C_(16)内引物巢式扩增后,12份标本中全部检出HBVDNA。说明巢式多聚酶链反应在乙型重症肝炎的病原诊断中,具有重要意义。
Severe cases of hepatitis B virus, the level of serum virus is so low that even in some cases even the most sensitive polymerase chain reaction is difficult to detect. Nested polymerase chain reaction can detect as low as 1-10 template DNA levels, we use this method to detect serum HBV DNA in 12 cases of severe hepatitis patients. Results: Four cases of HBVDNA were detected by C_1 and C_3 primers at one time, of which three cases were positive for HBeAg. After nested amplification of C_ (12) and C_ (16) primers, All detected HBVDNA. Nested polymerase chain reaction in the diagnosis of pathogen B severe hepatitis, is of great significance.
