目的:建立测定人血浆中头孢吡肟浓度的方法。方法:采用固相萃取(SPE)血浆中头孢吡肟。色谱柱为大连依利特Hypersil BDS C18,流动相为甲醇-磷酸盐缓冲液(20∶80),柱温为30℃,流速为1 ml.min-1,检测波长为257 nm,进样量20μl。结果:头孢吡肟检测浓度线性范围为1.0~200 mg.L-1(r=0.999 9);最低检测限为1 mg.L-1;低、中、高3种浓度头孢吡肟的方法回收率分别为118.0%,97.0%,95.0%,日内RSD分别为1.7%,3.6%,2.1%,日间RSD分别为8.2%,4.9%,2.6%。结论:该方法简便、灵敏度高,适用于测定人血浆中头孢吡肟的含量。
Objective: To establish a method for the determination of cefepime in human plasma. Methods: Cefepime in plasma was extracted by solid-phase extraction (SPE). The column was Dalian ELITE Hypersil BDS C18, the mobile phase was methanol-phosphate buffer (20:80), the column temperature was 30 ℃, the flow rate was 1 ml.min-1, the detection wavelength was 257 nm and the injection volume was 20μl . Results: The linear range of the concentration of cefepime was 1.0-200 mg.L-1 (r = 0.999 9), the lowest limit of detection was 1 mg.L-1, and the low, middle and high concentrations of cefepime were recovered The intraday RSD were 1.7%, 3.6% and 2.1%, respectively, and the intraday RSDs were 8.2%, 4.9% and 2.6% respectively, with rates of 118.0%, 97.0% and 95.0%, respectively. Conclusion: The method is simple, sensitive and suitable for the determination of cefepime in human plasma.
