目的研究在下颌骨发育过度导致的骨性Ⅲ类错畸形患者中,胰岛素样生长因子-1(IGF-1)基因的表达,探讨IGF-1基因表达与下颌骨生长的关系。方法选取骨性Ⅲ类错畸形患者27例,骨性Ⅰ类患者(包括个别正常)27例,应用荧光定量PCR技术和酶联免疫吸附剂测定(ELISA)技术研究下颌骨中IGF-1基因的表达变化。结果 RT-PCR检测结果显示,骨性Ⅲ类错畸形组中IGF-1 mRNA的表达量为(5.541 0±2.044 7)μg/μl,骨性Ⅰ类组中IGF-1 mRNA的表达量为(1.282 1±0.273 1)μg/μl,2组比较具有统计学差异(P<0.05)。根据IGF-1基因光密度值与浓度关系曲线,得回归系数0.998。ELISA检测结果显示,骨性Ⅲ类错畸形组中IGF-1蛋白浓度为(84.125 9±29.294 7)ng/L,骨性Ⅰ类组中IGF-1蛋白浓度为(22.406 4±4.931 2)ng/L,2组相比具有显著差异(P<0.05)。结论下颌骨生长量越大,IGF-1基因mRNA和蛋白表达越高。
Objective To investigate the expression of insulin-like growth factor-1 (IGF-1) gene in patients with skeletal Class Ⅲ malocclusion caused by overexpression of mandibular growth and to explore the relationship between IGF-1 gene expression and mandibular growth. Methods Twenty-seven patients with skeletal Class Ⅲ malocclusion and 27 patients with skeletal Class Ⅰ patients (including individual cases) were enrolled in this study. The expression of IGF-1 in mandibular bone was examined by fluorescence quantitative PCR and enzyme-linked immunosorbent assay (ELISA) Gene expression changes. Results The results of RT-PCR showed that the expression of IGF-1 mRNA in skeletal class Ⅲ malocclusion group was (5.541 ± 2.044 7) μg / μl, while that in skeletal class Ⅰ group was (1.282 1 ± 0.273 1) μg / μl, the difference between the two groups was statistically significant (P <0.05). According to the relationship between the optical density value and concentration of IGF-1 gene, the regression coefficient was 0.998. The results of ELISA showed that the IGF-1 protein level in skeletal Class Ⅲ malocclusion group was (84.125 9 ± 29.294 7) ng / L, IGF-1 protein level in skeletal Class I group was (22.406 4 ± 4.931 2) ng / L, there was a significant difference between the two groups (P <0.05). Conclusion The greater the growth of mandible, the higher the mRNA and protein expression of IGF-1.
