目的:研究天智颗粒对慢性脑缺血模型大鼠学习记忆能力和神经元特异性烯醇化酶(NSE)表达的影响。方法:采用双侧颈总动脉结扎方法建立慢性脑缺血模型,造模后60 d,采用三等分Y型电迷宫测定大鼠学习记忆能力,剔除不符合标准的动物。用天智颗粒(5 g.kg-1)治疗30d后采用三等分Y型电迷宫和免疫组化方法,观察比较各组大鼠行为学和NSE表达的差异。结果:治疗30d后,与模型组相比,治疗组学习记忆能力显著增加(P<0.01),假手术组和对照组大鼠皮质神经元无明显NSE表达,模型组NSE表达明显,天智颗粒治疗组NSE表达较模型组减弱,P<0.01。结论:天智颗粒可以减弱慢性脑缺血诱发的NSE表达,这是天智颗粒治疗血管性痴呆和慢性脑缺血的一种可能机制。
Objective: To study the effects of Tianzhi Granule on learning and memory ability and neuron-specific enolase (NSE) expression in rats with chronic cerebral ischemia. METHODS: Chronic cerebral ischemia models were established using bilateral common carotid artery ligation methods. Sixty days after modeling, the three-divided Y-type electric maze was used to determine the learning and memory ability of rats and to eliminate animals that did not meet the standard. After 30 days of treatment with Tianzhi Granules (5 g.kg-1), three quarters of Y-type electric maze and immunohistochemistry were used to observe the differences in behavior and NSE expression between the rats in each group. Results: After 30 days of treatment, the learning and memory ability of the treatment group was significantly increased compared with the model group (P<0.01). There was no obvious expression of NSE in the sham-operated group and the control group. The expression of NSE in the model group was obvious. The expression of NSE in the group was decreased compared with the model group, P<0.01. CONCLUSION: Tianzhi granules can attenuate the expression of NSE induced by chronic cerebral ischemia. This is a possible mechanism of Tianzhi granules in the treatment of vascular dementia and chronic cerebral ischemia.
