目的:研究金属硫蛋白对活性氧诱导的血管平滑肌细胞增殖的影响。方法:用过氧化氢和甲萘醌分别作为内源性和外源性活性氧刺激平滑肌细胞增殖,应用细胞总蛋白定量,3HTdR 参入,细胞周期时相测定等方法测定细胞增殖。结果:过氧化氢(25 μmol·L- 1)和甲萘醌(0 .5 μmol·L- 1) 可诱导培养的大鼠主动脉平滑肌细胞增殖,使其蛋白总量增加,S期细胞比例增高,DNA合成率增加。金属硫蛋白可以抑制过氧化氢和甲萘醌诱导的血管平滑肌细胞增殖。结论:金属硫蛋白对预防由活性氧诱导的血管平滑肌细胞增殖所引起的疾病可能具有重要作用。
Objective: To investigate the effect of metallothionein on the proliferation of vascular smooth muscle cells induced by reactive oxygen species. Methods: Hydrogen peroxide and menaquinone were used as endogenous and exogenous reactive oxygen species to stimulate smooth muscle cell proliferation. Cell proliferation was determined by total cellular protein quantification, 3H-TdR incorporation and cell cycle phase assay. Results: Hydrogen peroxide (25 μmol·L-1) and menadione (0.5 μmol·L-1) induced the proliferation of aortic smooth muscle cells in rat aortic smooth muscle cells and increased the total protein content. The proportion of S phase cells Increased, DNA synthesis rate increased. Metallothionein can inhibit the proliferation of vascular smooth muscle cells induced by hydrogen peroxide and menadione. CONCLUSIONS: Metallothionein may play an important role in the prevention of diseases caused by reactive oxygen species-induced proliferation of vascular smooth muscle cells.
