目的研究宁波市男男同性恋(MSM)人群中HIV-1感染者的免疫状况和HIV毒株的亚型分布特点。方法收集宁波市13名已经被确认为HIV-1型阳性的MSM的全血样品,对CD4+T淋巴细胞进行绝对数计数,运用套式PCR方法扩增病毒膜蛋白env基因的C2-V3区并进行序列测定。应用DNASTAR软件对序列和参考序列进行比对分析,确定基因亚型。结果成功获得11条env基因序列,确定1株为B亚型,4株为01_AE流行重组型,6株为07_BC流行重组型。该11例HIV-1阳性全血的CD4+T淋巴细胞绝对数最低106个/μl,最高649个/μl,平均350个/μl。结论本研究表明,宁波市MSM人群中存在多种HIV-1亚型,近半数的感染者免疫状况低下需要抗病毒治疗,流行形势严峻,应加强对该人群免疫状况和HIV-1毒株亚型变异的监测,及时调整防治策略。
Objective To study the immunological status of HIV-1 infected people and the distribution of subtypes of HIV strains in male and female homosexuality (MSM) population in Ningbo. Methods Thirteen HIV-1-positive MSM samples collected from Ningbo City were collected and the absolute numbers of CD4 + T lymphocytes were counted. The C2-V3 region of the viral envelope protein env gene was amplified by nested PCR And sequenced. DNASTAR software was used to compare the sequences and reference sequences to determine the genotype. Results Eleven env gene sequences were successfully obtained. One strain was identified as subtype B, four strains were 01_AE epidemic and six strains were 07_BC epidemic. The absolute number of CD4 + T lymphocytes in the 11 HIV-1 positive whole blood was the lowest 106 / μl, the highest 649 / μl with an average of 350 cells / μl. Conclusions This study shows that there are many HIV-1 subtypes in MSM population in Ningbo. Nearly half of those infected need anti-virus treatment because of their low immune status. The epidemic situation is grim. The immune status of this population and the prevalence of HIV-1 strain Type variation monitoring, timely adjustment of prevention and control strategies.
