目的 :研究白介素 - 2 (interleukin- 2 ,IL - 2 )基因转染的人卵巢癌细胞对人外周血淋巴细胞 (peripheral bloodiymphocytes,PBL)杀瘤活性的影响。方法 :采用脂质体携带 IL- 2基因转染人卵巢癌细胞系 3AO细胞 ,与正常人 PBL 混合培养 ,利用 MTT法测定混合培养后 PBL对已转染 IL- 2基因的 3AO细胞抑制率。结果 :基因转染后 3AO细胞生长良好 ,光镜形态下未观察到凋亡改变 ;转染后的 3AO细胞转录并产生 IL - 2 m RNA;PBL对转基因 3AO细胞的抑制率明显高于无基因转染的 3AO细胞的抑制率 (P<0 .0 1)。结论 :IL- 2基因修饰的卵巢癌细胞能够改变淋巴细胞的功能而发挥其抗肿瘤作用。
Objective: To investigate the effect of interleukin - 2 (IL - 2) gene transfected human ovarian cancer cells on the cytotoxicity of human peripheral blood lymphocytes (PBL). Methods: Human ovarian cancer cell line 3AO was transfected with IL-2 gene by liposome and mixed with normal human PBL. The inhibitory rate of PBL on 3AO cells transfected with IL-2 gene was determined by MTT assay. Results: 3AO cells grew well after transfection, and no apoptotic changes were observed under light microscope. Transfection of 3AO cells resulted in transcription of IL - 2 m RNA. The inhibition rate of PBL on transgenic 3AO cells was significantly higher than that of non - gene The inhibition rate of transfected 3AO cells (P <0.01). Conclusion: IL-2 gene-modified ovarian cancer cells can change the function of lymphocytes to exert their anti-tumor effect.
