三氧化二砷(As_2O_3)诱导人肺腺癌GLC-82细胞凋亡是一个多基因调控过程,不同时相基因表达存在明显差异,有些基因经As_2O_3诱导后持续表达或瞬时表达,有些基因经As_2O_3作用后表达水平降低或完全被抑制。对其中的6个差异片断进行了回收、克隆及Northern杂交验证。经序列测定,并与GenBank已知序列进行同源性比较后发现:wa1与wa2为新的基因片断;wa4与wb9均与rRNA的部分序列高度同源;wb182与线粒体细胞色素C氧化酶Ⅰ亚单位基因、wb162与humcox17R基因有高度的同源性。wb182与wb162均在As_2O_3诱导后24h表达明显降低。上述结果表明,线粒体细胞色素C氧化酶基因及humcox17R基因的低表达可能参与了As_2O_3诱导的人肺腺癌细胞凋亡的调节。
Arsenic trioxide (As_2O_3) induces apoptosis of human lung adenocarcinoma GLC-82 cells is a multi-gene regulatory process, there are significant differences in the expression of different phase genes, some genes are persistently expressed or transiently expressed after being induced by As 2 O 3 , and some genes are expressed by As 2 O 3 . The level is reduced or completely suppressed. Six of these differential fragments were recovered, cloned and verified by Northern hybridization. After sequence comparison and comparison with known sequences in GenBank, we found that wa1 and wa2 are novel gene fragments; wa4 and wb9 are highly homologous to partial sequences of rRNA; wb182 and mitochondrial cytochrome C oxidase I subunits The unit gene, wb162, has a high degree of homology with the humcox17R gene. Both wb182 and wb162 were significantly reduced at 24 h after induction of As 2 O 3 . The above results indicate that the low expression of mitochondrial cytochrome C oxidase and humcox17R genes may be involved in the regulation of As 2 O 3 -induced apoptosis of human lung adenocarcinoma cells.
