茜草炭不同提取物对子宫出血模型大鼠影响的比较研究

目的:观察茜草炭不同提取物对子宫出血模型大鼠的影响,筛选茜草炭止血作用的最佳提取方式。方法:空白组与其余各组分别灌胃给予0.5%CMC-Na、戊酸雌二醇(2.5 mg·kg~(-1)),连续灌胃3周,以宫血宁为阳性药,对照组及模型组静脉滴注羧甲基纤维素钠(CMC-Na),其他各组静脉滴注给予相应的治疗药物,观察茜草炭醇提物、复合提取物、醇提后水提物以及水煎物对子宫出血模型大鼠血液流变学、血小板系统以及纤溶系统的影响。结果:与空白组比较,模型组大鼠低切变率下的全血黏度降低(P<0.05),TT、PT、APTT显著延长(P<0.05、0.01),FIB、6-keto-PGF1α含量显著增加(P<0.01),TXB2和6-keto-PGF1α/TXB2显著降低(P<0.05、0.01)。与模型组比较,茜草炭复合提取物可显著升高各切变率下全血黏度(P<0.05、0.01),水煎物显著提高低切变率下全血黏度(P<0.05);醇提物、复合提取物和醇提后水提物显著缩短TT、PT、APTT(P<0.05、0.01),水煎物显著降低APTT(P<0.05),醇提物和复合提取物显著增加FIB含量(P<0.05),复合提取物和醇提后水提物显著降低6-keto-PGF1α含量(P<0.05、0.01),显著升高TXB2含量(P<0.05),复合提取物显著升高6-keto-PGF1α/TXB2比值(P<0.05),复合提取物有效缓解子宫内膜增厚、出血现象。结论:茜草炭复合提取物止血作用强于茜草炭其他提取物,即复合提取法为茜草炭止血作用的最佳提取方式。 OBJECTIVE: To observe the effects of different extracts of Rubia Cordifolia on uterine bleeding model rats, and to screen the best extraction method for the hemostatic effect of Rubia cordifolia. Methods: 0.5% CMC-Na and estradiol valerate (2.5 mg · kg -1) were administered orally in the blank group and the other groups respectively. The rats in the blank group were given gavage for 3 weeks continuously. The rats in the model group were treated with sodium carboxymethylcellulose (CMC-Na) by intravenous drip. The other groups were treated with intravenous drip and the corresponding therapeutic drugs were given to the mice in the model group. Results: Compared with the blank group, the whole blood viscosity of the model group was significantly decreased at low shear rate (P <0.05), TT, PT and APTT were significantly prolonged (P <0.05,0.01), FIB and 6-keto-PGF1α (P <0.01), TXB2 and 6-keto-PGF1α / TXB2 decreased significantly (P <0.05, 0.01). Compared with the model group, the extract of Rubia cordifolia could significantly increase the whole blood viscosity at each shear rate (P <0.05, 0.01), and the decoction significantly increased the whole blood viscosity at low shear rate (P <0.05) Extract, compound extract and alcohol extract significantly reduced TT, PT and APTT (P <0.05, 0.01), and decoction significantly decreased APTT (P <0.05). Alcohol extract and compound extract significantly increased FIB (P <0.05). The combined extracts and alcoholic aqueous extract significantly reduced the content of 6-keto-PGF1α (P <0.05, 0.01), significantly increased TXB2 content (P <0.05) 6-keto-PGF1α / TXB2 ratio (P <0.05), the compound extract effectively alleviate the endometrial thickening, bleeding. Conclusion: The hemostatic effect of compound extract of Rubia cordifolia is stronger than that of other extracts of Rubia cordifolia, that is to say, the compound extraction method is the best way to extract the hemostatic effect of Rubia cordifolia.
