利用液 质联用技术 (LC ESI MS)对啤酒酵母甘露聚糖乙酰解产物进行分析。电喷雾离子源为Na+ ;利用氨基柱对甘露聚糖乙酰解产物进行分离 ;流动相为乙腈 水 (70∶30 ,体积比 )。结果表明 ,该甘露聚糖的侧链是由甘露糖、甘露二糖、甘露三糖和甘露四糖组成。将该方法用于低聚糖相对分子质量的测定 ,结果准确、可靠、快速。
The beer yeast mannan acetyl hydrolyzate was analyzed by liquid chromatography-mass spectrometry (LC ESI MS). The electrospray ionization source is Na +; the amino-acid column is used to separate the mannan acetyl hydrolyzate; the mobile phase is acetonitrile-water (70:30, volume ratio). The results show that the mannan side chains are composed of mannose, mannobiose, mannotriose and mannotetraose. The method is applied to the determination of relative molecular weight of oligosaccharides, the result is accurate, reliable and rapid.
