目的:将目标起始密码子多态性分子标记(SCoT)技术应用于铁皮石斛的遗传多样性研究。方法:采用SCoT分子标记技术和非加权平均距离法(UPGMA)对20份不同来源的人工栽培铁皮石斛样品进行遗传多样性分析。结果:从备选的56个引物中筛选出20个SCoT引物,对共扩增共获得226个条带,平均每个引物产生11.3个条带,其中多态性条带204条,多态性比率达到90.3%。结论:本实验发现不同来源的铁皮石斛人工栽培种存在较大的遗传差异,目前药材市场中的人工栽培铁皮石斛遗传多样性丰富,SCoT技术可有效应用于铁皮石斛的遗传多样性研究。
Objective: To study the genetic diversity of Dendrobium candidum by using the target start codon polymorphism molecular marker (SCoT) technique. Methods: The genetic diversity of 20 cultivated Dendrobium officinale samples from 20 different sources was analyzed by SCoT molecular markers and non-weighted average distance method (UPGMA). Results: A total of 226 SCOT primers were screened out from 56 candidate primers. A total of 226 bands were obtained by co-amplification, with an average of 11.3 bands per primer, of which 204 were polymorphic bands. Polymorphism The rate of 90.3%. Conclusion: The present study found that different sources of Dendrobium officinale cultivars there is a large genetic difference, the current medicinal herbs market Dendrobium candidum rich genetic diversity, SCoT technology can be effectively applied to the genetic diversity of Dendrobium candidum.
