目的 研究转GM—CSF及CD—80基因的EL—4细胞的体外生物学特性。方法 通过逆转录病毒载体,将小鼠GM—CSF及CD—80基因分别转入EL—4细胞中,经克隆筛选、建立转基因细胞库。结果 转基因肿瘤细胞在体外培养中的形态、生长速度及集落形成能力与其亲代细胞基本相同,转基因肿瘤细胞表面MHCI类及MHC Ⅱ类抗原表达与亲代细胞相同,仅转CD—80基因的肿瘤细胞诱发了CD—80的高表达,EL—4/GM—CSF细胞的GM—CSF表达量为24ng/10~6细胞/24小时。结论 本实验为制备转基因肿瘤疫苗打下可靠的基础。
Objective To study the in vitro biological characteristics of EL-4 cells transfected with GM-CSF and CD-80 genes. Methods The mouse GM-CSF and CD-80 genes were transfected into EL-4 cells by retroviral vector and cloned to establish the transgenic cell bank. Results The morphology, growth rate and colony-forming ability of transgenic tumor cells in vitro were basically the same as those of their parental cells. The expression of MHC class I and MHC class II on the surface of transgenic tumor cells was the same as that of their parental cells. The expression of CD-80 was high, and the GM-CSF expression level of EL-4 / GM-CSF cells was 24 ng / 10 to 6 cells / 24 hours. Conclusion This experiment laid a solid foundation for the preparation of transgenic tumor vaccine.
