探讨阿魏酸钠对TNF α致血管内皮细胞 (VEC)中Bcl 2和Bax表达改变的影响及其细胞保护作用。以人脐静脉内皮细胞 (HUVEC)为实验模型 ,采用细胞培养、电镜、免疫组化等技术观察TNF α对VEC的形态以及内皮细胞中Bcl 2和Bax表达的影响 ,并研究阿魏酸钠的保护机理。结果 ,TNF α能明显损伤VEC的超微结构 ,可见较多凋亡细胞 ,细胞中Bcl 2表达明显减少 ,Bax表达明显增多。将阿魏酸钠与TNF α共同作用于VEC ,可见内皮细胞的超微结构与正常细胞形态相似 ,Bcl 2表达增多 ,Bax表达减少。阿魏酸钠可减少TNF α导致内皮细胞的损伤和凋亡 ,其机理可能是通过减少Bax表达 ,提高Bcl 2表达实现的
To investigate the effect of sodium ferulate on the expression of Bcl 2 and Bax in vascular endothelial cells (VEC) induced by TNFα and its cytoprotective effect. Using human umbilical vein endothelial cells (HUVEC) as an experimental model, cell culture, electron microscopy, and immunohistochemistry were used to observe the effect of TNFα on the morphology of VECs and the expression of Bcl 2 and Bax in endothelial cells, and to study the effects of sodium ferulate. Protection mechanism. RESULTS: TNFα significantly impaired the ultrastructure of VECs, and more apoptotic cells were seen. The expression of Bcl 2 was significantly decreased and the expression of Bax was significantly increased. The combination of sodium ferulate and TNFα in VEC shows that the ultrastructure of endothelial cells is similar to that of normal cells, and the expression of Bcl 2 is increased and that of Bax is decreased. Sodium ferulate can reduce the damage and apoptosis of vascular endothelial cells induced by TNFα. The mechanism may be through reducing the expression of Bax and increasing the expression of Bcl 2 .
