目的:研究不同浓度当归贝母苦参丸含药血清抑制胃癌细胞SGC-7901的机制。方法:将120只Wistar大鼠分为当归贝母苦参丸高、中、低剂量组(0.3,0.2,0.1 g·kg~(-1))和正常组,每组30只,ig 2次/d,连续给药7 d,制备含药血清,以空白血清处理胃癌细胞SGC-7901为空白组,实验组以当归贝母苦参丸含药血清高、中、低剂量组处理SGC-7901细胞后用RTq PCR技术检测环氧合酶-2(COX-2),Bcl-2相关X蛋白(Bax),人第10号染色体缺失的磷酸酶(PTEN)mRNA表达水平、用免疫细胞化学技术检测COX-2,BAX,PTEN蛋白表达情况。结果:与正常组比较,高剂量组使SGC-7901细胞COX-2 mRNA的表达量下降68%,BAX和PTEN mRNA的表达量分别升高99.7%和85.6%(P<0.05),含药血清可以降低SGC-7901细胞COX-2的阳性表达率,升高其平均灰度值,升高BAX和PTEN的阳性表达率,减低其平均灰度值(P<0.05)。结论:当归贝母苦参丸含药血清能抑制胃癌细胞SGC-7901,BAX,PTEN,COX-2 mRNA和蛋白的表达。
Objective: To study the mechanism of different concentrations of Radix Angelica Sinensis Medicinal serum inhibiting gastric cancer cell line SGC-7901. Methods: One hundred and twenty Wistar rats were divided into three groups: high, medium and low doses (0.3, 0.2, 0.1 g · kg -1) and normal group / d for 7 days. The serum containing SGC-7901 was treated with blank serum. The experimental group was treated with high, medium and low dose of SGC-7901 The expression of cyclooxygenase-2 (COX-2), Bcl-2 related protein X (Bax) and phosphatase (PTEN) mRNA on human chromosome 10 were detected by RT- Detection of COX-2, BAX, PTEN protein expression. Results: Compared with the normal group, the expression of COX-2 mRNA in SGC-7901 cells decreased by 68% and that of BAX and PTEN mRNA increased by 99.7% and 85.6% (P <0.05) The positive expression rate of COX-2 in SGC-7901 cells was decreased, the average gray value was increased, the positive expression rates of BAX and PTEN were increased, and the average gray value was decreased (P <0.05). Conclusion: Angelica sinensis, Radix Salviae miltiorrhized Pill can inhibit the expression of SGC-7901, BAX, PTEN and COX-2 mRNA and protein in gastric cancer cells.
