目的探讨T细胞免疫球蛋白黏蛋白-3(Tim-3)及其配体半乳凝素-9(Gal-9)与子宫内膜癌病理分级和分期的相关性。方法选取2012年6月-2014年6月42例子宫内膜癌患者组织样本为研究对象(观察组),其中低分化癌13例,中分化癌14例,高分化癌15例;Ⅰ期9例,Ⅱ期11例,Ⅲ期12例,Ⅳ期10例。选取20例子宫内膜单纯性增生患者子宫内膜组织样本为对照组。采用Western blot检测Tim-3及Gal-9蛋白表达水平;采用RT-PCR检测Tim-3及Gal-9 mRNA表达水平。结果 Western blot和RT-PCR结果显示:观察组Tim-3及Gal-9蛋白和mRNA表达水平均明显高于对照组(P<0.05),不同病理分级的子宫内膜癌组织中的Tim-3及Gal-9蛋白和mRNA表达水平依次为低分化>中分化>高分化(P<0.05);不同临床分期的子宫内膜癌组织中的Tim-3及Gal-9蛋白和mRNA表达水平依次为Ⅳ期>Ⅲ期>Ⅱ期>Ⅰ期(P<0.05)。结论 Tim-3及Gal-9蛋白和mRNA表达水平与子宫内膜癌的病理分级和分期具有相关性,Tim-3及Gal-9可作为子宫内膜癌临床诊断及预后的重要指标。
Objective To investigate the relationship between T-cell immunoglobulin mucin-3 (Tim-3) and its ligand Gal-9 and the pathological grading and staging of endometrial carcinoma. Methods Forty-two patients with endometrial cancer from June 2012 to June 2014 were selected as study group (observation group), including 13 cases of poorly differentiated carcinoma, 14 cases of moderately differentiated carcinoma and 15 cases of well differentiated carcinoma. Cases, 11 cases of stage Ⅱ, 12 cases of stage Ⅲ, 10 cases of stage Ⅳ. 20 cases of endometrial simple hyperplasia patients endometrial tissue samples for the control group. The expression of Tim-3 and Gal-9 protein was detected by Western blot. The expression of Tim-3 and Gal-9 mRNA was detected by RT-PCR. Results The results of Western blot and RT-PCR showed that the expressions of Tim-3 and Gal-9 protein and mRNA in the observation group were significantly higher than those in the control group (P <0.05). The expression of Tim-3 in different pathological grades of endometrial carcinoma (P <0.05). The expression levels of protein and mRNA of Tim-3 and Gal-9 in different clinical stages of endometrial carcinoma were Stage Ⅳ> stage Ⅲ> stage Ⅱ> stage Ⅰ (P <0.05). Conclusion The expression of Tim-3 and Gal-9 protein and mRNA are correlated with the pathological grade and stage of endometrial carcinoma. Tim-3 and Gal-9 may serve as important markers in clinical diagnosis and prognosis of endometrial carcinoma.
