目的比较同患者恶性胸腔积液上清、白细胞层及离心细胞块三种不同成分DNA提取质量,探索最有效的胸腔积液成分用于DNA提取及方法。方法收集解放军第一七五医院2014年细胞学及细胞块明确诊断恶性新鲜胸腔积液5例,分别用胸腔积液上清、白细胞层及离心细胞块三种不同成分进行DNA提取,然后对DNA含量和纯度进行测定,比较其DNA提取效果。结果对同一患者胸腔积液中白细胞层提取DNA含量最高,纯度次之,但对后续的EGFR基因突变和测序状况无影响;细胞块DNA提取质量次之,但纯度最高;胸腔积液上清提取质量及纯度均最低。结论恶性胸腔积液DNA提取用于EGFR基因突变检测的最佳成份为离心白细胞层,离心细胞块次之,本实验组胸腔积液上清提取效率不高。
Objective To compare the DNA extraction quality of three different components of malignant pleural effusion supernatant, white blood cell layer and centrifugal cell mass in patients with malignant pleural effusion to explore the most effective pleural effusion components for DNA extraction and methods. Methods Five cases of malignant fresh pleural effusion diagnosed by cytology and cell mass were collected from the 175th Hospital of People’s Liberation Army in 2014. DNA was extracted from three different components, pleural effusion supernatant, leukocyte layer and centrifugal cell mass respectively. Content and purity were measured to compare the DNA extraction. Results The DNA content of the leukocyte layer in pleural effusion of the same patient was the highest, followed by the purity, but had no effect on the follow-up mutation and sequencing of EGFR gene. The quality of cell DNA extraction was the second, but the purity was the highest. The pleural effusion supernatant The lowest quality and purity. Conclusions The best component of DNA extraction of malignant pleural effusion for EGFR gene mutation detection is centrifugal leukocyte layer, followed by centrifugal cell block. The efficiency of extraction of pleural effusion supernatant in this experimental group is not high.
