目的:比较异波帕胺(Ibo)与哇巴因(Oua)正性肌力作用药效的动力学。方法:以离体猫右心室乳头状肌递增药浓观察正肌效应。最大反应是药物递增到发生自发收缩前的反应。以lg[E/E_(max)— E]对lg C得Hill方程。结果:Hill方程中S值:Ibo及Oua分别为1.16与1.97,Ibo的S值小于Oua,使C_(95)/C_5(产生95%及5%最大药效的药浓)Ibo(264)是Oua(22.9)的11倍。与Oua相比:Ibo起效浓度低,斜率小及最大药效的浓度高。药浓下降时,Ibo的药效-药浓曲线呈现逆时针滞后襻,比Oua更明显。结论:在药浓增加及下降时,Ibo的正肌效应的变化比Oua为小。
OBJECTIVE: To compare the kinetics of the inotropic effects of ibo and ouabain (Oua). Methods: The positive muscle effect was observed by increasing the drug concentration in the left ventricular papillary muscles of isolated cats. The biggest reaction is the drug increases to occur before spontaneous contraction of the reaction. The equation lg for lg [E / E_ (max) - E] gives Hill’s equation. Results: The S value of Hill equation: Ibo and Oua were 1.16 and 1.97, respectively, and the S value of Ibo was less than that of Oua. The value of C 95 / C 5 (drug concentration producing 95% and 5% Oua (22.9) 11 times. Compared to Oua: Ibo has low onset of action, high slope, and maximum potency. When the drug concentration is declining, Ibo’s potency-drug concentration curve exhibits a counter-clockwise lag, which is more pronounced than Oua. CONCLUSION: The positive muscle effect of Ibo changes less than Oua at increasing and decreasing drug concentration.
