目的:研究药材巴戟天提取物(寡糖Ⅰ)的糖、蛋白质含量以及微量元素的分析。方法:采用苯酚-硫酸法测定总寡糖的含量,考马斯亮蓝染色法测定蛋白质含量,ICP-AES法分析其中的微量元素。结果:该提取物含总寡糖量为89.4%,总提取物中蛋白质含量为0.19%,经过D-941阴离子树脂柱层析富集脱蛋白后未测出含有蛋白质,微量元素测定其含有Zn、Fe、Ca等十二种微量元素,未检测出As、Cd、Pb等有害元素。结论:该提取物中含具有抗抑郁作用的高纯度植物寡糖。
Objective: To study the analysis of sugar, protein content and trace elements in the medicinal material Morinda officinalis (oligosaccharide I). Methods: The content of total oligosaccharides was determined by phenol-sulfuric acid method, protein content was determined by Coomassie blue staining, and trace elements were analyzed by ICP-AES. Results: The extract contains 89.4% total oligosaccharides and 0.19% protein in total extracts. Proteins were not detected after deproteinization by D-941 anion resin column chromatography, and trace elements contained Zn. Twelve kinds of trace elements such as Fe, Ca, and other harmful elements such as As, Cd, and Pb were not detected. Conclusion: This extract contains high purity plant oligosaccharides with antidepressant effect.
