目的:采用响应曲面法以蔗糖为指标优化北沙参中糖类的提取工艺条件。方法:首先进行单因素试验,分别考察乙醇浓度、提取时间和水料比三因素对蔗糖提取率的影响,在此基础上,进行响应曲面分析,优化北沙参蔗糖的最佳工艺条件。结果:最佳提取工艺条件为乙醇浓度39%、提取时间27 min、水料比57:1,在此条件下,可获得实际平均蔗糖提取率达6.02%。结论:优化工艺稳定可靠,可为中试生产北沙参提供理论依据。
OBJECTIVE: To optimize the extraction conditions of sugars in Radix isatidis by using response surface methodology with sucrose as an index. Methods: Firstly, single factor experiments were conducted to study the effect of ethanol concentration, extraction time and water to material ratio on the sucrose extraction rate. On the basis of this, the response surface analysis was carried out to optimize the optimum technological conditions of sucrose. Results: The optimal extraction conditions were as follows: ethanol concentration 39%, extraction time 27 min, ratio of water to material 57: 1. Under this condition, the average extraction rate of sucrose was 6.02%. Conclusion: The optimized process is stable and reliable, which can provide the theoretical basis for the production of Beiha.
