目的 :探讨 P5 3基因的变化与肿瘤细胞凋亡的关系。方法 :对两个分别具有野生型和突变型 P5 3基因的膀胱癌细胞株MGHU1和 RT112进行 8Gyγ射线照射 ,并采用定时显微录影设备观察 2 40 h,对每个被观察的细胞进行谱系分析。结果 :两个细胞株均出现了以凋亡为主的细胞死亡 ,累积细胞凋亡指数分别为 98和 10 5 .3,比较指数曲线后未发现显著性差异 ,P >0 .0 5。结论 :虽然两个细胞株的 P5 3基因状态和总 P5 3蛋白含量有所不同 ,但并没有影响 γ射线照射后的细胞凋亡的形成 ,从而未能影响细胞的放射敏感性。亦表明 P5 3基因对细胞凋亡的影响可能取决于不同的细胞类型。
Objective: To explore the relationship between the changes of P53 gene and tumor cell apoptosis. METHODS: Two bladder cancer cell lines, MGHU1 and RT112, with wild-type and mutant P53 gene, respectively, were irradiated with 8 Gy γ-rays, and the pedigrees of each observed cell were analyzed using a timed micro video recorder for 24 h. . Results: Apoptosis-dominated cell death occurred in both cell lines, and the cumulative apoptotic index was 98 and 105.3. No significant difference was found after comparing the exponential curves. P>0.05. Conclusion: Although the P5 3 gene status and total P53 protein content of the two cell lines are different, they did not affect the formation of apoptosis after γ-irradiation and did not affect the radiosensitivity of the cells. It also shows that the effect of P53 gene on apoptosis may depend on different cell types.
