目的:应用近红外光谱法和数据分析软件,对金银花中绿原酸含量进行快速测定。方法:利用HPLC测定样品中绿原酸的含量,运用偏最小二乘(PLS)法建立其含量与NIR光谱之间的多元校正模型,对未知样品进行含量预测。结果:建立的绿原酸校正模型相关系数(R2)、内部交叉验证均方差(RMSECV)、校正均方差(RMSEC)分别为0.993,0.380,0.158。经外部验证,校正模型的预测均方差(RMSEP)、平均回收率分别为0.170%,101.78%。结论:此方法具有快速简便,准确无损的特点,可以应用于金银花中绿原酸含量的快速检测。
Objective: To determine the content of chlorogenic acid in honeysuckle by near-infrared spectroscopy and data analysis software. Methods: The contents of chlorogenic acid in the samples were determined by HPLC. The multivariate calibration model between the contents and the NIR spectra was established by partial least squares (PLS) method, and the content of chlorogenic acid in the samples was predicted. Results: The correlation coefficients (R2), RMSECV and RMSEC of the chlorogenic acid calibration model were 0.993, 0.380 and 0.158 respectively. External validation showed that the mean square error (RMSEP) of the calibration model was 0.170% and the average recoveries were 101.78%. Conclusion: This method is fast, simple, accurate and non-destructive, and can be applied to the rapid detection of chlorogenic acid in honeysuckle.
