目的研究缺氧预适应小鼠脑匀浆提取液保护作用的机制。方法以SNAP为一氧化氮(NO)供体,应用反映细胞损伤程度的LDH透出率和反映细胞存活率的MTT比色法OD值为检测指标,观察NO对PC12细胞的毒性作用及缺氧预适应小鼠脑匀浆提取液对其毒性的影响。结果NO对PC12细胞可以产生剂量依赖性的直接毒性作用,而缺氧预适应小鼠脑匀浆提取液能有效对抗NO神经毒性作用。结论缺氧预适应脑组织对缺氧性神经元损伤具有保护作用的可能机制之一是通过抑制NO的神经毒性。
Objective To study the protective mechanism of brain homogenate extract from hypoxic preconditioned mice. Methods SNAP was used as a nitric oxide (NO) donor. LDH leakage rate and MTT colorimetric OD value which reflect the degree of cell damage were used as indexes to observe the toxic effects of NO on PC12 cells and hypoxia Effect of preconditioned mouse brain homogenate on its toxicity. Results NO produced a dose-dependent and direct toxic effect on PC12 cells. However, hypoxic preconditioning brain homogenate extract could effectively antagonize NO neurotoxicity. Conclusions One of the possible mechanisms of hypoxic preconditioning brain tissue protective effect on hypoxic neuronal injury is through inhibiting the neurotoxicity of NO.
