目的探讨信号转录和转录活化因子3(Signal transducer and activator transcription3,STAT3)在宫颈鳞状上皮病变组织中的表达及意义。方法采用RT-PCR方法检测13例宫颈炎组织、12例宫颈上皮内瘤变(Cervical intraepithelialneoplasia,CIN)Ⅰ级、17例CINⅡ-Ⅲ级及33例宫颈鳞状上皮细胞癌(squamous cell carcinoma,SCC)组织中STAT3mR-NA表达.结果 STAT3的表达与宫颈上皮病变的级别和临床分期无关(P均>0.10),与病理分级呈正相关(r=0.52,P<0.05),有脉管浸润和淋巴结转移组及浸润深度>3mm组显著高于无脉管浸润和淋巴结转移组及浸润深度≤3mm组,(P均<0.05);Cox回归分析结果显示STAT3的表达与宫颈癌的不良预后无关。结论 STAT3不能直接促进SCC的发生、发展,但对SCC的浸润、转移、分化起直接的促进作用,STAT3能否成为宫颈癌基因治疗的靶点有待于进一步研究。
Objective To investigate the expression and significance of Signal Transducer and Activator Transcription 3 (STAT3) in cervical squamous cell carcinoma. Methods Thirteen cases of cervicitis, 12 cases of grade Ⅰ cervical intraepithelial neoplasia (CIN), 17 cases of CINⅡ-Ⅲ grade and 33 cases of cervical squamous cell carcinoma (SCC) were detected by RT-PCR. ) STAT3mR-NA expression in the tissue.Results The expression of STAT3 was not related to the grade and clinical stage of cervical epithelial lesions (P> 0.10), and positively correlated with pathological grade (r = 0.52, P <0.05) Metastasis group and infiltration depth> 3mm group was significantly higher than those without vascular invasion and lymph node metastasis and infiltration depth ≤3mm group (P <0.05). Cox regression analysis showed that STAT3 expression had no relationship with poor prognosis of cervical cancer. Conclusions STAT3 can not directly promote the occurrence and development of SCC, but plays a direct role in the invasion, metastasis and differentiation of SCC. Whether STAT3 can be a target of cervical cancer gene therapy remains to be further studied.
