为了比较不同价态的金属-Salen配合物与DNA的作用,按文献方法,合成了以N,N’-双(2-羟基-1-萘甲醛)缩邻苯二胺这种Salen配体,Salen配体与醋酸锰(钴、铜、锌)4种金属盐配位形成的锰、钴2种3价金属配合物及铜,锌2种2价金属配合物,探究了它们与DNA之间的相互作用.2种3价配合物在紫外滴定DNA实验和粘度试验中均产生了明显作用效果;而2价金属配合物没有表现出明显的效果,DNA溶液的粘度也没有显示出与配合物浓度的正相关性.结果表明,3价配合物以插入模式与DNA结合,而且在与DNA相互作用上,Co(Ⅲ)的配合物略强于Mn(Ⅲ)的配合物;而2价金属配合物可能以其它模式与DNA作用,其并没有表现出经典的插入结合特征.
In order to compare the role of metal-Salen complexes and DNA in different valence states, the Salen ligands with N, N’-bis (2-hydroxy-1-naphthaldehyde) Salen ligand and manganese acetate (cobalt, copper, zinc) four kinds of metal salt coordination of manganese, cobalt two kinds of trivalent metal complexes and copper, zinc two kinds of bivalent metal complexes to explore their DNA and between .The two kinds of trivalent complexes have obvious effect in UV titration DNA test and viscosity test, while the bivalent metal complexes did not show obvious effect, the viscosity of DNA solution also did not show any significant interaction with the complex The results showed that the trivalent complex bound to DNA in the insertion mode and the complex of Co (Ⅲ) was slightly stronger than that of Mn (Ⅲ) in interaction with DNA. However, the bivalent metal Complexes may interact with DNA in other modes, which do not exhibit classical insertion-binding characteristics.
