[Objectives] The aim was to establish a UPLC method for simultaneous determination of two components in Erjing pills and compare the content changes of rutin and chlorogenic acid in the preparing process of Polygonatum sibiricum Delar. as well as optimize the compatibility of Erjing pills. [Methods] The chromatographic separation was carried out at 35 ° C on an ACQUITY UPLC HSS T3( 100 mm × 2. 1mm,1. 8 μm) column,the mobile phase was methanol( A) and 0. 2% phosphoric acid solution( B),the detective wavelength was 345 nm with the flow rate of 0. 4 m L / min. [Results]The contents of rutin and chlorogenic acid in Erjing pills after processing were 2- 3 times before processing. [Conclusions] The processed mixture of Polygonatum sibiricum Delar. and Lycium barbarum L. is the best,the method is simple,accurate and reproducible.
