目的 :进一步调查无血清培养基血清代用品中因子对体外无血清培养的人表皮细胞生长增殖的影响。方法 :利用L16(4 5)正交试验对无血清培养基血清代用品 (SerumSubstitute ,SS)中五因子的四种不同浓度进行调查 ,用MTT法测定它们分别的效应。结果 :SS中除Chol作用可疑外 ,BSA、Cata、Ins、TF均能明显促进表皮细胞的生长增殖 ,它们较好的组合是BSA 15mg/ml、Cata 10 μg/ml、Ins 8μg/ml、TF 5 μg/ml。结论 :SS在体外无血清培养基中能替代血清较好的促进表皮细胞的生长
OBJECTIVE: To further investigate the effects of serum-free medium serum substitutes on the growth and proliferation of human epidermal cells in serum-free culture in vitro. Methods: Four different concentrations of five factors in SerumSubstitute (SS) serum-free medium were investigated by L16 (4 5) orthogonal test, and their respective effects were measured by MTT assay. Results: In addition to the suspicion of Chol in SS, BSA, Cata, Ins and TF could significantly promote the proliferation of epidermal cells. The best combinations of them were BSA 15mg / ml, Cata 10μg / ml, Ins 8μg / ml, TF 5 μg / ml. Conclusion: SS can replace epidermal cell growth in serum-free medium in vitro
