目的观察贝那普利(Ben)与美托洛尔(Met)合用8wk、16wk对高血压大鼠动脉收缩压(SBP)、左心室肥厚和心肌纤维化的作用。方法采用两肾一夹Goldblat高血压模型;尾容积法测定清醒大鼠动脉收缩压;以左心室羟脯氨酸含量反映心肌纤维化程度。结果肾动脉术后12和20wk时模型组SBP,左心室重与体重之比(LVW/BW)和左心室胶原蛋白含量(LVCC)均高于假手术组;20wk时该3项指标的净增值分别为:ΔSBP为126,ΔLVW/BW为131,ΔLVCC为38。与模型组比,术后12wkBen15mg·kg-1·d-1,ig+Met10mg·kg-1·d-1,ig联合用药组的ΔSBP,ΔLVW/BW,ΔLVCC分别下降了78%、80%和78%;术后20wk时,上述指标分别下降87%、92%和90%;且低于两个单用药组。结论Ben和Met在抑制高血压大鼠及心肌纤维化方面有协同作用,这种作用随治疗时间的延长而明显。
Objective To observe the effects of combination of Ben and metoprolol on the systolic blood pressure (SBP), left ventricular hypertrophy and myocardial fibrosis in hypertensive rats. Methods Two kidneys and one clip of Goldblat model of hypertension were used. The tail volume method was used to measure the systolic pressure in awake rats. The level of hydroxyproline in left ventricle was measured to reflect the degree of myocardial fibrosis. Results The SBP, LVW / BW and LVCC of the model group were significantly higher than those of the sham operation group at 12 and 20wk after the renal artery surgery. The net value of the three indexes at 20wk Respectively: ΔSBP of 126, ΔLVW / BW of 131 and ΔLVCC of 38. Compared with the model group, the ΔSBP, ΔLVW / BW and ΔLVCC decreased by 78%, 80% and 78% respectively in the ig combined group after 12wkBen15mg · kg-1 · d-1 and ig + Met10mg · kg-1 · d- ; 20wk after surgery, the above indicators decreased by 87%, 92% and 90%; and lower than the two single-agent group. Conclusions Ben and Met have a synergistic effect in inhibiting hypertensive rats and myocardial fibrosis, and this effect is obvious with prolonged treatment time.
