目的:研究猫爪草总皂苷对H_(22)实体瘤的增殖及其自噬基因表达的影响。方法:昆明小鼠48只,随机分为4组:模型组、环磷酰胺阳性对照组及猫爪草总皂苷低、高剂量组,每组12只。采用小鼠腋下注射H_(22)细胞株建立实体瘤模型,造模同时给予相应药物进行干预3 w。末次给药后记录瘤质量指数和胸腺、脾脏、肝脏、肾脏脏器系数、HE染色检测肿瘤组织病理学变化,并采用实时荧光定量PCR检测肾组织中凋亡基因BAX、BCL-2和自噬相关基因Atg5、Beclin1的表达水平。结果:猫爪草总皂苷可显著降低H_(22)实体瘤质量系数,抑瘤率达38.6%对肿瘤细胞具有明显的杀伤作用,并能显著促进BAX mRNA的表达,抑制BCL-2 mRNA的表达;并能上调Atg5、Beclin1 mRNA的表达。结论:猫爪草总皂苷可以抑制H_(22)肝肿瘤的增殖,其机制与促进肿瘤细胞的凋亡和自噬活动有关。
Objective: To investigate the effects of Radix Ranunculus total saponins on the proliferation of H_ (22) solid tumors and the expression of autophagy genes. Methods: Forty eight Kunming mice were randomly divided into 4 groups: model group, cyclophosphamide positive control group and low and high dose group, 12 rabbits in each group. The model of solid tumor was established by injecting H22 cell line into the armpit of mice. The model was given with the corresponding drugs for 3 weeks. The index of tumor mass, the index of thymus, spleen, liver and kidney were recorded after the last administration, the pathological changes of tumor were detected by HE staining, and the apoptosis gene BAX, BCL-2 and autophagy were detected by real-time fluorescence quantitative PCR Related genes Atg5, Beclin1 expression levels. Results: The total saponin of Catharanthus rubra could significantly reduce the mass index of solid tumor of H_ (22), and the inhibition rate of tumor reached 38.6%. It could significantly promote the expression of BAX mRNA and inhibit the expression of BCL-2 mRNA ; And can up-regulate Atg5, Beclin1 mRNA expression. Conclusion: The total saponins of Radix Ranunculus could inhibit the proliferation of H_ (22) hepatocarcinoma, the mechanism of which is related to the promotion of tumor cell apoptosis and autophagy.
