目的观察大鼠内皮抑素cDNA联合反义血管内皮生长因子(VEGF)核苷酸对胶质瘤生长的影响。方法利用经抗性筛选可表达内皮抑素的C_6细胞(C_6/endo)、有效转染有反义VEGF164核苷酸片段的C_6细胞(C_6/VEGF~-)及转染有可表达内皮抑素和反义VEGF154核苷酸片段的C_6细胞(C_6/endo-VEGF~-)裸鼠皮下注射制作胶质瘤模型,21 d后测定移植瘤瘤体和瘤重,ELISA法检测肿瘤组织中的VEGF含量,免疫组织化学方法检测肿瘤组织的微血管密度。结果C_6/endo、C_6/VEGF~-和C_6endo-VEGF~-的胶质瘤组织生长明显较C_6胶质瘤慢(P<0.01或P<0.05),但C_6/endo和C_6/endo-VEGF~-生长差异无统计学意义,前三者VEGF含量也明显低于后者,差异有统计学意义(P<0.01),其中C_6/endo-VEGF~-的VEGF含量低于C_6/endo、C_6/VEGF~-(P<0.01);C_6/endo和C_6/endo-VEGF~-的血管密度较C_6/VEGF~-低(P<0.05),但是前两者间差异无统计学意义。结论C_6endo和C_6endo-VEGF~-可通过下调VEGF来达到抑制肿瘤生长;C_6/endo- VEGF~-抗肿瘤生长更多可能是依赖于内皮抑素。
