目的探讨研究蛋白磷酸酶I(protein phosphatase1 regulatory subunit 3,PPP1R3)在直肠癌SW480细胞中的黏附、运动、迁移和侵袭能力的生物学活性。方法用PPP1R3、EGFR抗体、VEGFR抗体干预SW480细胞后,分别对其行细胞运动实验、黏附实验、迁移及侵袭实验,分析PPP1R3对SW480细胞运动、黏附、迁移和侵袭能力的影响及其作用机制。结果随着PPP1R3浓度的增加,直肠癌SW480细胞的黏附、运动、迁移和侵袭能力明显减退(P<0.05)。而且穿过人工基底膜的能力明显减低(P<0.05)。结论 PPP1R3能够抑制直肠癌SW480体外的侵袭、转移作用,可为PPP1R3基因表达缺失的直肠癌术后患者提供放化疗、生物靶向治疗依据。
Objective To investigate the biological activity of protein phosphatase 1 regulatory subunit 3 (PPP1R3) in adhesion, motility, migration and invasion in rectal cancer SW480 cells. Methods After SW480 cells were treated with PPP1R3, EGFR and VEGFR antibodies, the effects of PPP1R3 on the motility, adhesion, migration and invasion of SW480 cells and their mechanism were analyzed by cell migration assay, adhesion assay, migration and invasion assay. Results With the increase of PPP1R3 concentration, the adhesion, motility, migration and invasion of rectal cancer SW480 cells were significantly reduced (P <0.05). And the ability to cross the artificial basement membrane was significantly reduced (P <0.05). Conclusion PPP1R3 can inhibit the invasion and metastasis of rectal cancer SW480 in vitro and provide chemoradiotherapy for patients with postoperative rectal cancer with deletion of PPP1R3 gene.
