目的研究中医复合因素造模法塑造脾虚型大鼠模型的效果,并探讨中医复合因素造模的特点和优势。方法采用中医传统病因:饥饱失常+劳倦+破气耗气方法塑造脾气虚型大鼠模型,将30只大鼠随机分为正常组15只,模型组15只,模型组予以每日灌服小承气汤溶液(剂量为2.5g/kg),隔日半量进食,每日下午将大鼠放入水深50cm水温20℃的水槽中游泳,以力竭为度(大鼠鼻尖没入水面10s),造模2周。正常组每日正常饮食活动,而后两组大鼠每日灌服生理盐水(剂量为5ml/kg),连续灌胃15d,通过观察大鼠的一般表现,体重、进食、饮水量、游泳力竭时间、胃排空、胃黏膜情况评价脾虚型大鼠模型的效果。结果造模后模型组大鼠的体重、进食、饮水量以及游泳力竭时间均下降,与正常组比较差异具有统计学意义,(P<0.05或0.01),经过灌胃生理盐水15d后,模型组大鼠与正常组大鼠在体重、进食、饮水量以及游泳力竭时间差异仍有统计学意义,且胃排空率明显低于正常组,(P<0.01)。结论中医复合因素能较快塑造脾虚型大鼠的证候,并且效果可维持,简便,价格低廉,更符合中医证候的内涵。
Objective To study the effect of TCM composite factor modeling in shaping rat model of spleen deficiency syndrome and to explore the characteristics and advantages of TCM compound factor modeling. Methods Traditional Chinese etiology: Familial insufficiency + fatigue + gas-broken gas method was used to model spleen-deficiency rats. Thirty rats were randomly divided into normal group (n = 15) and model group (n = 15) Xiaochengqi Decoction solution (2.5g / kg), half a day to eat the next day the rats into the water depth of 50cm water temperature of 20 ℃ swimming in the pool, to exhaustion degree (rat nose tip into the water 10s) Modeling 2 weeks. The rats in the normal group were fed with normal diet daily. The rats in the two groups were fed with normal saline (5ml / kg) daily for 15 days. The rats’ general performance, body weight, food intake, water intake and swimming exhaustion Time, gastric emptying, gastric mucosal evaluation of the effect of spleen deficiency rat model. Results After model establishment, the body weight, food intake, water intake and swimming exhaustion time of model group rats decreased. Compared with normal group, the difference was statistically significant (P <0.05 or 0.01). After intragastric administration of saline for 15 days, There were still significant differences in body weight, food intake, water intake and swimming exhaustion time among rats in normal group and rats in normal group, and the rate of gastric emptying was significantly lower than that in normal group (P <0.01). Conclusion TCM composite factors can quickly shape the syndrome of spleen deficiency rats, and the effect can be maintained, simple, cheap, more in line with the connotation of TCM syndromes.
