[目的]探讨大蒜油对胃癌SGC7901细胞增殖的抑制作用。[方法]①胃腺癌SGC7901细胞在体外常规培养,给予不同浓度大蒜油处理,观察细胞生长情况,用四唑盐(MTT)比色法检测细胞增殖,计算抑制率。②细胞给予大蒜油或转化生长因子α(TGF-α)处理,或同时给予大蒜油和TGF-α处理,用MTT比色法检测细胞增殖情况,计算抑制率。[结果]①胃癌SGC7901细胞常规培养,加入5%、10%、15%大蒜油作用72 h时,其细胞增殖明显受抑制,抑制率分别为24.5%、44.0%和54.8%。②加入2.5%低浓度大蒜油作用72 h,与2.5%血清培养比较,细胞增殖速度无明显变化;加入30μg/L TGF-α时,细胞增殖明显加快;当同时加入2.5%大蒜油和30μg/L TGF-α时,则TGF-α的促增殖作用明显受抑制,抑制率为18.1%。[结论]大蒜油能够抑制胃癌细胞增殖,并能抑制TGF-α刺激的胃癌细胞增殖。提示大蒜油可能对胃癌细胞的TGF-α自分泌、旁分泌促增殖环路具有抑制作用。这可能是大蒜油抗肿瘤细胞增殖的作用方式之一。
[Objective] To investigate the inhibitory effect of garlic oil on the proliferation of gastric cancer SGC7901 cells. [Method] ① The gastric adenocarcinoma SGC7901 cells were cultured in vitro and treated with different concentrations of garlic oil. The growth of SGC7901 cells was observed. The proliferation of SGC7901 cells was detected by MTT colorimetric assay. The cells were treated with garlic oil or transforming growth factor-α (TGF-α) or garlic oil and TGF-α at the same time. The cell proliferation was detected by MTT assay and the inhibition rate was calculated. [Results] ① The proliferation of SGC7901 cells was inhibited by adding 5%, 10% and 15% garlic oil for 72 h. The inhibitory rates were 24.5%, 44.0% and 54.8%, respectively. ②Compared with 2.5% serum, the cell proliferation rate had no significant change after adding 2.5% low concentration garlic oil for 72 h; when 30 μg / L TGF-α was added, the cell proliferation was obviously accelerated; when adding 2.5% garlic oil and 30 μg / L TGF-α, then the role of TGF-α-induced proliferation was significantly inhibited, the inhibition rate was 18.1%. [Conclusion] Garlic oil can inhibit the proliferation of gastric cancer cells and inhibit the proliferation of gastric cancer cells stimulated by TGF-α. Tip garlic oil may gastric cancer cells TGF-α autocrine, paracrine promote proliferation cycle has an inhibitory effect. This may be one of the ways garlic oil anti-tumor cell proliferation.
