目的:探讨干扰素调节因子-4结合蛋白(interferon regulatory factor-4 binding protein,IBP)对人涎腺腺样囊性癌(salivary adenoid cystic carcinoma,SACC)细胞侵袭能力和上皮间质转化的影响.方法:采用Transwell试验,将穿膜细胞进行染色计数并分析各组细胞侵袭能力的变化,明确IBP对细胞侵袭能力的影响;通过波形蛋白和角蛋白的免疫荧光染色,观察比较ACC2-C1/IBP和ACC2-C1细胞的染色强度.结果:ACC2-C1/IBP细胞侵袭率大于ACC2-C1细胞和ACC2细胞;在波形蛋白中的染色 ACC2-C1/IBP 细胞强于 ACC2-C1细胞,在角蛋白中的染色ACC2-C1/IBP细胞弱于ACC2-C1细胞.结论:IBP有促进SACC细胞侵袭的作用;初步证实了IBP能够促进人涎腺腺样囊性癌细胞上皮间质转化的发生.“,”Objective: To investigate the effect of interferon regulatory factor-4 binding protein(IBP)on invasion and epithelial-mesenchymal transition in salivary adenoid cystic carcinoma cells.Methods:The ability of cell invasion was based on analysis of stained-cells penetrating study by Transwell experiments to determine the effects of IBP on the ability of cell invasion.The staining intensity of ACC2-C1/IBP and ACC2-C1 cells was observed and compared by the vimentin and ck8 immunofluorescence stain.Results:The invasion rate of ACC2-C1/IBP cells was higher than that of ACC2-C1 cells and ACC2 cells.The staining of ACC2-C1/IBP cells in vimentin was stronger than that in ACC2-C1 cells,and the staining of ACC2-C1/IBP cells in ck8 was weaker than that in ACC2-C1 cells.Conclusion:IBP can promote SACC cells invasion,and it was preliminary confirmed that IBP can promote the occurrence of epithelial-mesenchymal transition.
