参附益心颗粒对大鼠急性心梗后心功能及心肌组织的影响

目的:观察参附益心颗粒对急性心梗后大鼠心功能及心肌组织病理演变的影响。方法:雄性SD大鼠,行冠状动脉左前降支结扎术制备大鼠急性心肌梗死模型,将制备成功者随机分为模型组、参附益心临床等效剂量组(1.76g/kg)及氯沙坦阳性对照组(10 mg/kg),假手术组仅开胸并在左前降支下过线。分2批分别灌胃给药1周/4周后,采用彩色多普勒超声影像系统监测各组大鼠超声心电图,分析测量EF、FS、LVIDD及LVIDS等反映心功能的指标;并取心室肌组织制备石蜡包埋组织切片,行HE染色及Masson染色,镜下观察并拍照。通过纵向和横向比较探讨参附益心颗粒对急性心梗后大鼠心功能及心肌组织病理演变的影响。结果:急性心梗后参附益心颗粒(1.76g/kg)给药1周,与模型组相比仅EF值有明显改善,病理学检测显示心梗组织主要表现为心肌细胞的死亡和早期的炎症反应如炎细胞浸润、血管增生及成纤维细胞增生,参附益心颗粒可起到保护心肌细胞及抑制炎症反应的作用;心梗后4周,与心梗后1周相比,心功能指标表现为恶化趋势,但参附益心组(1.76g/kg)与模型组同期相比EF、FS及LVIDS均有明显改善,病理结果显示心梗组织表现为室壁变薄及组织的纤维化,交界区纤维化组织呈树枝样向正常组织渗透性生长,参附益心颗粒(1.76g/kg)可对心肌组织的纤维化起到一定的抑制作用,使心肌纤维化程度和范围有所控制。结论:参附益心颗粒可改善心功能,对急性心梗后组织可起到保护心肌和抑制其纤维化的作用,从而阻滞和延缓心梗后慢性心衰的发生和发展。 Objective: To observe the effect of Shenfu Yixin Granule on cardiac function and myocardial histopathological changes in rats after acute myocardial infarction. Methods: Acute myocardial infarction (AMI) model was established in male Sprague-Dawley rats by ligating left anterior descending coronary artery. The rats were randomly divided into model group, reference clinical effective dose group (1.76g / kg) and chlorine Sham-positive control group (10 mg / kg), sham-operated group only thoracotomy and in the left anterior descending branch under the line. The rats were divided into 2 groups by intragastric administration for 1 week / 4 weeks respectively. Paraffin-embedded tissue sections were prepared from muscle tissue, HE staining and Masson staining were performed, observed under the microscope and photographed. The effects of Shenfuyixin Granule on cardiac function and myocardial histopathological changes in rats after acute myocardial infarction were compared by vertical and horizontal comparison. Results: Acute myocardial infarction after Shenfu Granule (1.76g / kg) for 1 week, compared with the model group EF only significantly improved, pathological examination showed myocardial infarction mainly as myocardial cell death and early Inflammatory reactions such as inflammatory cell infiltration, vascular proliferation and fibroblast proliferation, Shen Fuyi heart particles can play a protective role of myocardial cells and inhibit the inflammatory response; 4 weeks after myocardial infarction, compared with 1 week after myocardial infarction, heart The functional indexes showed a worsening tendency, but the parameters of Fuganyixin group (1.76g / kg) were significantly improved compared with the model group, EF, FS and LVIDS, pathological results showed that the myocardial infarction showed thinning of the wall and tissue Fibrosis, junctional fibrosis tissue was like to the normal tissue osmotic growth, Shen Fuyi heart particles (1.76g / kg) can play a certain role in the inhibition of myocardial fibrosis, the degree and extent of myocardial fibrosis Controlled. Conclusion: Shenfu Yixin Granule can improve cardiac function, and play an important role in protecting myocardium and inhibiting its fibrosis after acute myocardial infarction, thus blocking and delaying the occurrence and development of chronic heart failure after myocardial infarction.
