目的确定双泽清脂颗粒罐组逆流提取的产业化生产工艺。方法采用单因素及正交试验的方法,以大黄酚的转移率和浸膏得率的加权得分为综合评价指标,考察每罐每次提取加水量、提取时间、提取溶剂温度及混合药材粉碎粒度对提取工艺的综合影响,确定最佳工艺,并通过中试放大,考察工艺的稳定性,并与原工艺进行对比研究。结果最佳工艺为混合药材粉碎成中粉粒度、3罐逆流提取,每罐每次加入12倍量水、提取溶剂保持在70℃提取,每次提取40min。结论罐组式逆流提取技术应用于本品的提取工艺中具有显著性优势,优选的工艺参数稳定可行。
Objective To determine the industrial production process of countercurrent extraction of Shuangze Qingzhi granules tank. Methods The single factor and orthogonal test methods were used to evaluate the transfer rate of chrysophanol and the weighted score of the extract yield as the comprehensive evaluation index. The extraction water quantity, extraction time, extraction solvent temperature and granulation size The comprehensive impact on the extraction process to determine the best technology, and enlarged through the pilot test to study the stability of the process, and compared with the original process. Results The best process was that the mixed medicinal materials were crushed into medium particle size, 3 cans were countercurrently extracted, 12 times the amount of water was added into each can and the extraction solvent was kept at 70 ℃ for extraction for 40min. Conclusion Tank group countercurrent extraction technology applied to the extraction of this product has a significant advantage, the preferred process parameters are stable and feasible.
