目的:观察与相同剂量的单药相比,褪黑素(MT)与酵母硒(Se)联合应用是否能提高对肿瘤的治疗效果及其机制。方法:将S180肉瘤细胞接种于小鼠右腋皮下,分为7组:模型组、Se 100μg.kg-1组、Se200μg.kg-1组、MT40 mg.kg-1组、MT80 mg.kg-1组、Se 100μg.kg-1+MT40 mg.kg-1组、环磷酰胺(CTX)组。10 d后处死小鼠。测定肿瘤组织内细胞周期蛋白依赖性激酶2(CDK2)和p53的表达水平。结果:MT与Se联合应用可显著提高抑瘤率,降低肿瘤重量,降低CDK2的表达水平,提高p53和p21的表达水平。结论:与相同剂量的单药治疗肿瘤相比,MT与Se两者联合应用能明显提高疗效。
OBJECTIVE: To observe whether the combination of melatonin (MT) and yeast selenium (Se) can improve the therapeutic effect and mechanism of tumors compared with the same dose of a single drug. METHODS: S180 sarcoma cells were inoculated into the right iliac crest of mice and divided into 7 groups: model group, Se 100 μg.kg-1 group, Se200 μg.kg-1 group, MT40 mg.kg-1 group, MT80 mg.kg- One group, Se 100μg.kg-1+MT40 mg.kg-1 group, cyclophosphamide (CTX) group. Mice were sacrificed 10 days later. The expression levels of cyclin-dependent kinase 2 (CDK2) and p53 in tumor tissues were measured. RESULTS: The combination of MT and Se could significantly increase the tumor inhibition rate, reduce tumor weight, decrease the expression of CDK2, and increase the expression of p53 and p21. Conclusion: Compared with the same dose of monotherapy for tumors, the combination of MT and Se can significantly improve the efficacy.
