用体外长期培养的方法建立了小鼠胎肝基质细胞系(murinefetallivercel,MFLC),并运用免疫荧光技术、电镜技术、倍增时间的测定及染色体检查等方法,对此细胞系进行了细胞学鉴定。此细胞系建立至今已2年,历经3个阶段。细胞为多角形,用免疫荧光法显示胞浆内含角蛋白,同时也表达S-100和波形蛋白。电镜观察可见细胞形态多样,有些细胞含成束的张力丝,有些相邻细胞间有桥粒结构,部分细胞内可见病毒颗粒。细胞Ia表达呈强阳性,细胞倍增时间为24h。染色体为异倍体(50~78条),结果表明,本实验所建成的MFLC是以上皮细胞为主的已转化的胎肝混合细胞系。
Murinefetallivercel (MFLC) was established by long-term culture in vitro and the cell lines were identified by immunofluorescence, electron microscopy, doubling time and chromosome examination. This cell line has been established for 2 years, after three stages. The cells were polygonal, showing cytoplasmic keratin by immunofluorescence as well as S-100 and vimentin. Electron microscopy shows that the cells are diverse in morphology. Some cells contain bundles of tension filaments. Some of the adjacent cells have desmosomes and some of the cells can be seen virus particles. Cell Ia expression was strongly positive, cell doubling time was 24h. Chromosome aneuploid (50 ~ 78), the results show that the MFLC formed in this experiment is based on epithelial cells transformed fetal liver mixed cell line.
