目的介绍一种石蜡包埋细胞芯片的简易制备方法及验证其应用价值。方法收集科研培养细胞或临床细胞学标本,手工法制作石蜡包埋的细胞芯片,并进行HE染色、免疫组织化学法及TUNEL检测法验证细胞芯片的应用价值。结果此法熟练掌握后能制备出60个点阵左右的细胞芯片,肉眼观察点阵完整,排列均一,经HE染色、免疫组化染色及TUNEL凋亡染色证实,染色效果良好,结果可靠。结论简易手工法制备的石蜡包埋细胞芯片的操作容易,设备简单,能够迅速成型制备出多种不同类型的细胞芯片,值得推广应用。
Objective To introduce a simple preparation method of paraffin embedded cell chip and to verify its application value. Methods Cell culture or clinical cytology specimens were collected and paraffin-embedded cells were prepared by manual method. HE staining, immunohistochemistry and TUNEL were used to verify the value of the cell-based microarrays. The results of this method can master the preparation of about 60 dot matrix cell chip, the naked eye to observe the lattice integrity, arranged in uniform, by HE staining, immunohistochemical staining and TUNEL apoptosis staining confirmed that the staining effect is good, the result is reliable. Conclusion The paraffin embedded cell chip prepared by simple manual method is easy to operate and has simple equipment and rapid prototyping and preparation of many different types of cell chips, which is worth popularizing and applying.
