目的·针对泛耐药肺炎克雷伯菌寻找宽宿主谱噬菌体,并对其生物学特性、基因组成进行分析。方法·以泛耐药肺炎克雷伯菌为宿主菌,从医院污水中分离纯化得到针对宿主菌的泛耐药肺炎克雷伯菌噬菌体。电子显微镜观察噬菌体的大小和形态,分析其效价、宿主谱、酸碱稳定性和热稳定性。抽取噬菌体DNA进行测序及分析。结果·分离到l株肺炎克雷伯菌噬菌体,命名为SH-Kp152234。电子显微镜观察显示其属短尾噬菌体,基因组为线状双链DNA,全长为40 578 bp,GC含量为52.85%。预测共有49个开放阅读框,保守序列具有已知的相关功能。结论·筛选得到肺炎克雷伯菌噬菌体SH-Kp152234的宿主谱较宽,潜伏期较短,可适应不同的温度和p H值,为治疗泛耐药肺炎克雷伯菌感染提供潜在依据。
OBJECTIVE · To search broad host spectrum bacteriophage against pan-resistant Klebsiella pneumoniae and to analyze its biological characteristics and gene composition. Methods · Pan-drug-resistant Klebsiella pneumoniae as the host bacteria, isolated from the hospital sewage purification obtained against the host bacteria Pan-resistant Klebsiella pneumoniae phage. The size and morphology of the phage were observed under an electron microscope, and its potency, host spectrum, pH stability and thermal stability were analyzed. Phage DNA was extracted for sequencing and analysis. Results · Strain of Klebsiella pneumoniae phage was isolated and named SH-Kp152234. Electron microscopy showed that it belongs to the bacteriophage tail phage. The genome was linear double-stranded DNA with a total length of 40 578 bp and a GC content of 52.85%. A total of 49 open reading frames are predicted, and conserved sequences have known related functions. Conclusion · The Klebsiella pneumoniae phage SH-Kp152234 has a broad host spectrum and a short incubation period, and can be adapted to different temperatures and p H values. It provides a potential basis for the treatment of pan-drug-resistant Klebsiella pneumoniae infection.
